From car experience they will measure anything that may increase performance or reliability against official papers with dimensions etc as supplied by MV with the homologation papers for the bike:
Bore/Stroke, valve sizes/weights, piston shape/size/weight, intake size, butterfly size (including looking for knife edging etc) in fact anything that has beed altered from a production engine that is outside the rules
They tend to home in on the front running bikes etc and also bikes that may produce an unusual performance.
They watch lap times and speed trap readings as well as listening to other riders about other bikes performance
